Nutraceuticals and oxidative stress research

ACT4 technology supporting antioxidant research

Protecting active molecules and exploring new applications

Oxidative stress is a biological phenomenon associated with an imbalance between the production of oxidising species and the body’s antioxidant defences. Molecules that may modulate oxidative and inflammatory processes are an area of interest in nutraceutical development.

Many naturally occurring molecules have properties of interest in this context, but pose formulation challenges relating to poor water solubility, stability and bioavailability.

The ACT4 platform was developed to address these challenges by encapsulating lipophilic active ingredients in submicrometre emulsions. ACT4 is studying several molecules with the aim of exploring their potential through advanced formulations.

Curcumin: investigating antioxidant potential

Curcumin is a natural molecule of interest in research on oxidative and inflammatory processes. Its limited water solubility and bioavailability challenges make it a candidate for dedicated delivery systems.

ACT4 conducted a preclinical rat study to evaluate a formulation based on its technology platform. The results are described in the company blog and form part of its curcumin research programme.

Animal-model results are preliminary evidence. They cannot automatically be extrapolated to humans, but can help guide subsequent development.

Coenzyme Q10: from delivery to experimental cardioprotection

Coenzyme Q10 (CoQ10) is involved in the mitochondrial electron transport chain and antioxidant defence mechanisms, making it an ingredient of interest for nutraceutical applications.

ACT4 studied a CoQ10 formulation in an experimental model of cardiac injury associated with chemotherapy drugs, observing a cardioprotective effect under the conditions studied. In a further study in healthy mice, the formulation was associated with a reduction in some baseline inflammatory markers.

These are preclinical findings and currently do not demonstrate cardioprotective or anti-inflammatory efficacy in humans. They nevertheless provide a scientific basis for further evaluation of the formulation and its potential applications.

From preclinical testing to human development

ACT4 is working on the regulatory pathway for product registration with the Italian Ministry of Health, in preparation for subsequent clinical development and assessment of efficacy in humans.

The start of a clinical study will depend on completion of regulatory requirements and definition of the study protocol. The aim is to move from experimental results to rigorous assessment of safety, tolerability and efficacy in the population of interest.

Lycopene: exploring opportunities for cell protection

Lycopene is a lipophilic carotenoid whose antioxidant properties have been widely studied. Formulation and delivery are important technological aspects of its development.

ACT4 conducted an in vitro study to evaluate the potential cardioprotective effect of a lycopene formulation in a chemotherapy-associated cellular injury model. Described in the company blog, this is a preliminary cell-model investigation. Further work is needed to establish reproducibility and any biological or clinical relevance.

A technology platform for functional ingredients

ACT4’s interest extends beyond individual molecules. The platform addresses formulation challenges shared by lipophilic ingredients, with potential advantages for stability and integration into different products.

The research aims to build a scientific foundation for new nutraceutical ingredients, keeping experimental evaluation distinct from demonstrated clinical benefits in humans.
